Hi all, Just been reading the article about dissecting needles and wondered if anyone has any tips on preparing poroid fungi for microscopy i.e. soaking on liquid before to soften the fungus! I have looked on line for help but without success hence this request.

    Thanks on advance.

    Mike.

      Hi,

      I tend to leave most poroid’s alone – life’s too short! 😆

However, (although I haven’t used it myself), I understand that Clemenҫon’s solution is widely used to soften/rehydrate woody material (sometimes allied with mounting in ammoniacal solutions), prior to microscopy.

      See: –

for an explanation of the chemicals which are used to make-up Clemenҫon’s (and other) mycological reagents etc.

      Regards,

      Mike.

Brilliant, life may be too short but I am quite excited. Thanks for the information, really appreciate it.

      All the best,

      Mike.

I slice a piece from the bracket, and then use a pointy tweezer to pull off tiny strips. KOH solution seems to be a good way to soften tissue. I am sure there are better methods if you want really good photomicrographs, but it’s good enough to see features.

Thanks Leif, Time to get soaking and stripping. Hopefully these methods will glean some useful results.

      Mike.
