Polypore and crust preparation.

Hi all, Just been reading the article about dissecting needles and wondered if anyone has any tips on preparing poroid fungi for microscopy i.e. soaking on liquid before to soften the fungus! I have looked on line for help but without success hence this request.

Hi,

I tend to leave most poroid's alone - life's too short! :lol:

However, (although I haven't used it myself), I understand that Clemenҫon's solution is widely used to soften/rehydrate woody material (sometimes allied with mounting in ammoniacal solutions), prior to microscopy.

I slice a piece from the bracket, and then use a pointy tweezer to pull off tiny strips. KOH solution seems to be a good way to soften tissue. I am sure there are better methods if you want really good photomicrographs, but it's good enough to see features.
